- W1419666919 abstract "This chapter discusses the heterologous expression of G protein-linked receptors in pituitary and fibroblast cell lines. Receptors that couple with heterotrimeric G proteins have a characteristic heptahelical structure, and constitute a class of receptors for hundreds of endogenous amines, peptides, hormones, and odorants. For many of these receptors, particularly receptors for small molecules such as amines, the transmembrane domains dictate ligand binding specificity and provide the scaffolding for the three-dimensional structure of the receptor. The intracellular peptide loops, particularly those regions apposed to the plasma membrane, provide the contact points for specific noncovalent interactions that form the receptor–G protein ternary complex. On binding of the agonist to the receptor, an exchange of guanosine triphosphate (GTP) for guanosine diphosphate (GDP) at the G protein is catalyzed. Binding of GTP to the G protein, a subunit induces dissociation of the ternary complex, releasing an activated a subunit and βγ dimer that interact with various effector molecules to increase or decrease their activity. The application of molecular biology techniques such as low-stringency hybridization and polymerase chain reaction (PCR)-based cloning to the isolation of receptors, G proteins, and effectors has helped reveal the diversity and complexity of signal transduction systems." @default.
