• W1423707683 abstract "Schon seit einigen Jahren werden die Effekte von Stickstoffmonoxid (NO) als Mediator an den unterschiedlichsten Prozessen im Korper untersucht. Zunehmend ruckt der Einfluss von NO auf die Herzfrequenz neben dem ausgiebig untersuchten Effekt auf die myokardiale Kontraktion ins Interesse wissenschaftlicher Studien, wobei sowohl exogene NO-Donatoren als auch Modulatoren der endogenen NO-Synthase (eNOS) eingesetzt werden. Ziel der Untersuchung war es, an spontan schlagenden rechten Vorhofen von Mauseherzen(Stamm C57 black6) einerseits einen frequenzmodulierenden Effekt durch exogenes NO sowie den dafur verantwortlichen Mechanismus zu untersuchen, andererseits aber auch uber eine Modulation der endogenen NO-Produktion eine Beteiligung von NO an der Frequenzregulation am Sinusknoten nachzuweisen. Zusatzlich wurden der Einfluss von Bradykinin, einem Stimulator der endothelialen NO-Synthase , auf die Spontanfrequenz des Sinusknotens und der zugehorige Signaltransduktionsweg untersucht.Daruberhinaus wurde gepruft, ob der Betarezeptorenblocker Nebivolol, der die glatte Gefassmuskulatur uber eine Stimulation der NO-Produktion dilatieren soll, am Sinusknoten die Schrittmacheraktivitat steigert.Hierzu wurden die rechten Vorhofe in Organbader uberfuhrt,die spontanen Schalgfrequenzen registriert und ein steady state im Sinne einer konstanten Schlagfrequenz abgewartet. Dann wurden die Prufsubstanzen kumulativ bis zu den gewunschten Endkonzentrationen zugegeben und die so induzierten Frequenzanderungen registriert .Es liess sich ein positiv chronotroper Effekt durch exogen appliziertes NO nachweisen, welcher zum einen uber eine Aktivierung der loslichen Guanylatzyklase (sGC) und somit gesteigerter Produktion von cGMP, andererseits aber auch uber einen direkten Angriff am Schrittmacherkanal If vermittelt zu werden scheint.Der eNOS-Inhibitor L-NMMA fuhrte zu einer nur geringen Abnahme der Sinusknotenfrequenz, evtl. aufhgrund einer schon basal nur sehr geringen NO-Produktion, so dass die Inhibition nicht signifikant war. Bradykinin zeigte einen uber den Bradykinin-B2-Rezeptor vermittelten positiv chronotropen Effekt auf den Sinusknoten, wobei eine Stimulation der endogenen NO-Produktion uber die eNOS als auch eine Hemmung der Acetylcholin abhangigen Kaliumkanale eine Rolle zu spielen scheinen. Ein frequenzmodulierender Effekt von Nebivolol mit Beteiligung des NO-Signaltransduktionsweges konnte nicht nachgewiesen werden." @default.
