• W1429171507 abstract "Keratins constitute a family of approximately 20 related a-helix-rich structural proteins of 40-70 kDa (kilodaltons) and make up the subunits of the intermediate filaments present in all epithelial cells. This multigene family is divided into two distinct types on the basis of sequences that are predicted to form coiled coils: (1) the acidic keratins form type I and (2) the neutral-basic keratins form type II. Epithelial cells, in general, always express at least one member of each type and various factors influence which specific members are expressed such as the cell type, period of embryonic development, state of differentiation, and growth environment of the cell. However, the mechanisms regulating the expression of this family of genes are not known. To define the mechanism regulating keratin genes, it is necessary to have specific probes to monitor their expression at the molecular level. Therefore, cDNA clones corresponding to the major keratins synthesized in mouse epidermis have been isolated and characterized. Several lines of evidence are presented in the chapter that suggest that the expression of the subsets of keratin genes is coordinately regulated and dependent on the state of differentiation. In addition, on the basis of amino acid sequence information deduced from the nucleotide sequence of these cloned cDNAs, it is postulated that the sequence differences observed for keratins expressed at the different states of differentiation will change the properties and function of the filaments that they form." @default.
