- W1435424086 abstract "Introduction We have demonstrated in rat myocardial infarctions (MIs) that human umbilical cord blood mononuclear cells (HUCBC), which contain stem cells, reduce MI size by as much 85%, and increase LV EF by 35% compared with saline treated infarcts without requirements for rat immune suppression. We hypothesized that HUCBC increase cardiac endothelial cell and myocyte survival and limit ischemic by paracrine mechanisms. Methods We determined by microarray growth factor and anti-inflammatory cytokines secreted by HUCBC into culture media during hypoxia (1% O2, 94% N2, 5% CO2) for 12hrs (N=10). We then treated human coronary artery endothelial cell cultures and cardiac myocyte cultures with either acellular media taken from hypoxic preconditioned HUCBC cultures (N=10) or normal culture media (N=10) and subjected the endothelial and myocyte cultures to 24 hours of hypoxia or normoxia. We then determined endothelial and myocyte apoptosis by caspase and annexin determinations and cell necrosis by propidium iodide exclusion. Results Hypoxic stressed HUCBC increased secretion of hepatocyte growth factor by more than 375%, increased angiopoietin, angiogenin and VEGF more than 100–200%, and increased anti-inflammatory IL-4 and IL-10 more than 80% (p<0.01). HUCBC media reduced apoptosis of coronary artery endothelial cells and cardiac myocytes by 36 and 49% and decreased necrosis of these cells by 30 and 39% respectively (p<0.05). Conclusion HUCBC can protect ischemic myocardium by secreting biologically active growth factors and anti-inflammatory cytokines." @default.
