- W1437397725 abstract "Publisher Summary This chapter illustrates immunoelectron microscopy and image processing for epitome mapping. Immunoelectron microscopy (IEM) is a labeling method based on the analysis of immunocomplexes by electron microscopy (EM) that can be used to study a large variety of structures. However, specimen preparation and observation conditions (dehydration, exposure to the electron beam, etc.) may induce a broad range of deformations and partial destruction of biological molecules. In addition, the preliminary results obtained with specimens embedded in vitreous ice suggest that the negative stain could be replaced by frozen hydrated specimens. However, simple negative staining combined with a well-standardized method of image processing produces average images with a resolution of 3–4 nm. The topological location of epitopes refined by immunological mapping produces an epitope location that is surpassed only by the crystallographic determination of the structure of Fab-containing immunocomplexes by X-ray diffraction." @default.
