- W1438857552 abstract "Publisher Summary This chapter discusses the application of gas chromatography–mass spectrometry (GCMS) techniques to analyze prostaglandins and related substances. The GCMS technique is a powerful and sensitive technique that allows the detection of prostaglandins and related products in the picogram and femtogram range. This technique is applied to biological fluids, perfusates of isolated perfused organs, and to intact and disrupted cell incubates. It is potentially feasible to quantify the variety of fatty acids, mono-, di-and trihydroxy-fatty acids through to the prostaglandins and their metabolites and the dihydroxy leukotrienes and their w-hydroxy metabolites, all in one sample. Through the use of deuterium labeled precursor fatty acids are incorporated into the tissue lipids. It is possible to obtain information on the pool size of the precursor fatty acid and also to determine the specific activity (d/h) of the products newly formed by the tissue. The chapter presents examples t o illustrate the concept of profiling of the arachidonic acid cascade. The most attractive and promising future development in the use of mass spectrometry in the quantitative assay of products of biologic origin must be MSMS." @default.
