The monoclonal antibody libraries are used as a general approach to detect, classify, and investigate previously uncharacterized nuclear proteins. The chapter describes the production of monoclonal antibodies against specific proteins from an enriched but still heterogeneous nuclear fraction. Although most of the antibodies found are useful as molecular markers, it is inherent to the monoclonal library approach that at the beginning almost nothing is known about the proteins investigated. Most existing libraries are made using animal proteins, and much is undertaken in Drosophila melanogaster and several species of amphibia. Drosophila melanogaster provides an attractive system to which the monoclonal antibody library approach can be applied, in view of the possibility of carrying out cytological mapping of protein distribution on interphase polytene chromosomes. Recently, Mazzolini et al. have established 330 cell lines following immunization with crude nuclear extracts of Nicotiana tabacum. The cell lines they described secreted monoclonal antibodies to conserved histone H1 epitopes, and some of them detected HI in all higher eukaryotes.
