• W1444442364 abstract "Publisher Summary Under constant conditions, single-ion channels open and close in an apparently random manner. This chapter discusses the simplest model to observe this phenomenon in which there are two distinct states available to the channel: ions pass through if the channel is in the open state but not if it is in the closed state. If the two states are connected by first-order rate constants, the simple model scheme shown in the chapter predicts that the amount of time the channel stays open will follow an exponential distribution. The time constant of that exponential is also the mean open time of the channel, which equals the reciprocal of the rate constant for channel closing. The closed times will be distributed similarly. An earlier model for generation of the action potential in the squid giant axon remains the paradigm for the description of voltage-dependent conductance. The basis of this model is the existence of two separate pathways for the movement of Na + and K + ions across the membrane. Since the development of this model, voltage-clamp techniques have been used on dozens of cell types, and new techniques such as gating current and single-channel recording have given invaluable insight into channel gating. Several ion channel proteins have been cloned and sequenced, and molecular techniques are being used to study structure–function relations." @default.
