- W1446731708 abstract "This chapter provides an overview of electron microscopy of yeasts. Most electron microscope studies with yeasts are carried out on material fixed with permanganate. The fixation is often carried out in a 2% solution of KMnO4 in water for 1–2 hours at room temperature. In this simple procedure, fixation and staining are performed in one-step, and samples can be observed without any further staining. Poststaining with a uranyl salt (acetate or nitrate) either in the 70% step of the dehydration or on the section, and/or a lead salt such as lead citrate or lead oxide, enhances the contrast considerably. Cell walls and membranes are stained distinctly, but the nucleoplasm and the groundplasm often seem partly cleared. This is possibly due to oxidative degradation and dissolution of plasma components. Part of the contrast is because of the reaction of permanganate with unsaturated membrane lipids. It is assumed that the addition of permanganate to the double bond results in an anionic product, which moves to the plane of the polar groups. The fixation of whole-yeast cells with osmium tetroxide alone or after a preceding fixation with aldehyde does not give good results." @default.
