It considers the range of possible DNA and RNA analogs that have been developed and that can be envisaged as potential drugs. The chapter illustrates that some molecules are intended to act as drugs with two (or more) functional groups in which one is an oligomer analog and the other confers some specific property on the oligomer, such as improved cellular uptake or increased RNase activity as RNA is the target for the antisense approach. There is a great need of second-generation analogs that involve other substitutions and additions to the basic oligonucleotide structure but do not alter its conformation significantly, but also for third-generation analogs that are envisaged to involve wholesale alteration or replacement of the sugar-phosphate backbone. As penicillin analogs resulted in the maturing of antibiotic therapy in the 20th century, so oligomeric gene-mimetic analogs may be their 2lst-century equivalents." @default.
