- W1451133150 abstract "This chapter considers the expression and localization of keratins and the keratin-associated protein— filaggrin—during human fetal skin development from approximately 7 weeks estimated gestational age (EGA) until birth to approximately 40 weeks EGA. It reviews biochemical and immunological evidence for the time of expression of different members of the family of keratin proteins and filaggrin and correlates these changes with morphological changes during development. This review is useful for both basic and applied aspects of epidermal biology. The keratins are a large and complex family of proteins that make up the intermediate filaments in all epithelial cells, but only a limited number of keratins are expressed in any one type of epithelium or in the cells at a particular stage of differentiation of an epithelium. The correlation of biochemistry and morphology in developing fetal epidermis may give insight into the role of the specific members of the keratin family in cell function. Filaggrin is an unusual protein that aggregates in a stoichiometric manner with keratin filaments and is thought to function as the keratin matrix protein in the fully differentiated and cornified cells of epidermis. The interaction of these proteins may account for the so-called keratin pattern of negatively outlined filaments embedded in an electron-dense matrix. However, the initial keratinized cells formed during the human fetal development do not appear to have a keratin pattern. Thus, fetal development offers a situation in which the proposed role of filaggrin in keratinization can be investigated." @default.
