- W1454755217 abstract "This chapter discusses the determination of sorbitol dehydrogenase from spermatozoa. The reaction mixture in both the experimental and blank cuvettes (1-cm light path) contains 0.3 ml diphosphopyridine nucleotide (DPN), 0.3 ml Tris buffer, 0.3 ml sorbitol, and water to a final volume of 2.95 ml. The assay is run at room temperature. The reaction is started by addition of 0.05 ml enzyme to the experimental cuvette. The increase in absorbance at 340 mμis observed at 30-second intervals for 3 minutes. Ram semen is used as the starting material for the extraction of the enzyme. There are several accepted methods for the collection of ram semen. The one to be recommended depends on the use of the artificial vagina. Ram semen (about 10 ml) is diluted with 4 volumes of Ringer solution at room temperature, centrifuged at about 700 g for about 15 minutes. The supernatant liquid, which is the diluted seminal plasma, is discarded. The sedimented spermatozoa are washed three times, each time with approximately 10 ml of fresh Ringer solution; the last centrifugation has to be sharp (usually 15 minutes at 15,000 g in a Servall centrifuge) to obtain firmly packed spermatozoa and to keep the solvent retained by the pellet at a minimum. Data relating to the specificity of the sperm enzyme are listed along with those for the dehydrogenase from seminal vesicle." @default.
