As the specificity of these inhibitors is a major concern when using them with intact cells, their effects on isolated enzymes ­rather than on intact cells ­is the major focus of the chapter. Several approaches have been taken to find specific inhibitors of protein kinases. Many of the most interesting inhibitors have come from systematic screening of natural products, and these compounds have provided the basis for the synthesis of other more specific inhibitors. While a great deal of progress has been made and some very useful inhibitors have been discovered, the lack of specificity remains a problem. In the future, greater cognizance must be taken of the fact that cells contain a large number of protein kinases and that in vitro results with only a few enzymes cannot be extrapolated to the complex situation in an intact cell. There are several examples where the term “specific protein-kinase inhibitor” has proved to be a misnomer. The results with studies on the effects of protein-kinase inhibitors on intact cells often have rather malleable interpretations." @default.
