- W1458766504 abstract "Publisher Summary The need for innovative new drugs to treat central nervous system (CNS) disorders prompted the U.S. Congress to declare the 1990s as the “Decade of the Brain”. It has been a major initiative to facilitate the discovery and development of new medications not only to treat diseases of aging but also to develop new drugs for the treatment of anxiety, schizophrenia, epilepsy, pain, stroke, depression, and substance abuse. Despite an explosion of knowledge in the neurosciences, considerable investment from the pharmaceutical industry, and over 1200 products in the worldwide R & D pipeline in 1990, new drug introduction in the CNS area has not matched what has been seen in other therapeutic areas. Research in the area of animal behavior as related to human CNS disease states, both in terms of determining efficacy and side effect potential, has undergone a significant de-emphasis in the past decade as academia, government, and industry have focused considerable resources on molecular aspects of neuroscience. Animal models of CNS disorders are traditionally used to gain an indirect measure of compound penetration to the CNS. The relationship between the central nervous and immune systems has led to the concept of the immunologic brain. Immune system function is dependent on CNS viability, while many mediators of the immune system are present in the CNS and regulate neuronal function and viability. Despite the increased understanding of drug mechanisms at the molecular level, CNS drug discovery remains, in many respects, highly empirical. Significant changes in the health care and research environments, none unique to the brain, have set new guidelines for the choice of research projects within the industry and the challenges in compound advancement into and through the clinic." @default.
