- W1458778743 abstract "Publisher Summary This chapter discusses in vitro system whereby the characteristics of mammalian neuroendocrine cells identified by antidromic stimulation, dye-marking, and immunocytochemical and ultrastructural analysis can be correlated with hormone release. The chapter presents a method for analyzing vasopressin (AVP) supraoptic (NSO) magnocellular neuroendocrine cells (MgC) in the rat hypothalamo-neurohypophysial complex (HNC) in vitro . Rapidly removed from ventral hypothalamus, the HNC complex consists of the complete neuroendocrine final common pathway-supraoptic neurons, supraoptico-neurohypophysial tracts, and neural lobe. Under carefully controlled in vitro conditions of perifusion, the combination of electrophysiological studies of identified single vasopressin-containing neurons and stimulated AVP release are presented. The single-cell identification techniques include cell-marking with a fluorescent dye, immunocytochemical double-labeling, and diaminobenzidine photooxidation product (DAB-POP) triple-labeling for ultrastructural studies. The triple-labeling identification technique of neurons studied electrophysiologically, cell-marking with a fluorescent dye, immunocytochemical and ultrastructural analysis, may allow describing functional subsets of magnocellular peptidergic neurons more precisely." @default.
