- W1465684911 abstract "This chapter examines the synthesis and clinical studies of the antineoplastic agents. 5-Aziridinyl-2,4-dinitrobenamide has the exceptional activity against the Walker carcinoma. Its antitumor activity is reversed by 4-amino-5-imidazolecarboxamide. Suppression of cell membrane transport plays a role in the resistance to nitrogen mustard by the Yoshida sarcoma cells. Metabolism of cyclophosphamide by rat hepatic microsomes has been reported. Enzymatic phosphorylation studies confirm that ara-C and its phosphorylated derivatives are deoxycytidine antagonists rather than cytidine antagonists. Inhibition of DNA polymerase activity is considered more significant to antineoplastic properties than either blocking the uptake of thymidinel or the inhibition of the conversion of cytidine diphosphate to deoxycytidine diphosphate. 5-Fluorouracil (5-FU) might be more effective in patients with liver metastases when given orally, due to high drug concentrations in the portal system. Oral dosage with 5-FU is safe and effective in metastatic colorectal carcinoma and it enhances the antitumor effect of irradiation in bronchogenic carcinoma." @default.
