- W1468724118 abstract "This chapter discusses the androgen receptors in the rat ventral prostate and their hormonal control. The specific binding of androgens to their target organ receptors has been investigated mainly in the ventral prostate gland of the castrated adult rat. The use of sucrose or glycerol gradient ultracentrifugation has allowed the study of several properties of the cytoplasmic receptors. Short exposure of rat ventral prostate androstanolone–receptor complex to dextran-coated charcoal does not remove a large amount of nonspecific binding. Steroid hormone receptors are acidic proteins that can be precipitated selectively by protamine. An application of the principle of protamine sulfate precipitation to rat ventral prostate cytosol is described. The androstanolone-3H dpm precipitated in the presence of an excess androstanolone are subtracted from the androstanolone-3H dpm precipitated in the absence of excess androstanolone, and the difference is considered as representative of specific binding. It is reported that the β-protein, which is precipitated at 40% ammonium sulfate concentration, has an almost exclusive affinity for androstanolone whereas the α-protein, in addition to binding androstanolone, also binds testosterone, progesterone, and estradiol in significant amounts." @default.
