There are a number of enzyme-mediated labeling systems that utilize the lack of penetration of cells by large exogenously applied enzymes to label specific groups in the plasma membrane. Diazotized sulfanilic acid reagent reacts with histidine and tyrosine residues on membrane proteins in a mild reaction that occurs at neutral pH and is considered to have little effect on membrane function. Pyridoxal phosphate permeates some membranes very slowly and generates Schiff bases with protein amino groups, under physiological conditions, which are then tritiated by reduction with tritiated borohydride. Concanavalin A (Con A) is a sugar-specific lectin that binds to and agglutinates plant protoplasts. Lanthanum ions probably compete for calcium-binding sites and are known to penetrate the plasma membrane very slowly or not at all." @default.
