- W147179935 abstract "Publisher Summary This chapter focuses on developing systems for the malignant transformation of cultured liver cells. In the hamster embryo cell system, the cells are diploid at the time of treatment and transformation can be scored within 10 days. The nontransformed cells die after a few generations and cannot be cloned with high efficiency. Primary cultures of hamster embryonic cells are prepared from 12- to 14-day-old fetuses of Syrian hamsters. The malignancy of transformed colonies can be determined by isolating colonies from a treated dish and then growing up large numbers of cells derived from these colonies for injection into hamsters. The selection of suitable lines of mouse cells for in vitro chemical oncogenesis is of great importance. The cytotoxicity exerted by a chemical is related to its concentration and to the duration of the exposure. Dose–response curves for cytotoxicity should be done for each cell line and for each chemical. It is always necessary to correlate in vitro morphological transformation with in vivo malignancy. Before staining, some transformed foci are picked by the ring isolation technique and grown in culture." @default.
