It also describes alternative approaches to the formation of chimeric genes. The most direct approach involves utilizing two sets of uniquely matched restriction sites. When appropriate sites are not available where required, site directed mutagenesis could be used to design restriction sites that avoid altering the reading frame or individual residues. If an amino acid substitution cannot be avoided, the resulting protein must be evaluated. In another strategy, one fixed fusion module is incorporated into a transposon and used to generate chimeric genes. This has been developed for the evaluation of protein export and membrane topologies. In this approach, a DNA molecule has been designed to lack the promoter and translational start sequences of the structural gene of interest, and the 5'-truncated genetic cassette is inserted near the end of an IS (insertion sequence) element of a transposon carried on F factor." @default.
