- W1478012310 abstract "This chapter discusses glycoprotein membrane enzymes individually in a sequence concentrating initially on the enzymes of general distribution, followed by brush border hydrolases and acid hydrolases of the lysosome. The chapter concentrates on two aspects of glycoprotein membrane enzymes: (1) the characterization, membrane association, and glycoprotein nature of those enzymes that have been purified and (2) the controversial evidence regarding the degree of their localization at the external surface of the plasma membrane. The large number of glycoprotein ectoenzymes on the surface of the intestinal and renal brush borders makes them excellent organelles for studying enzyme–membrane relationships. All the enzymes are intrinsic membrane proteins in the sense that they are not easily extracted with salt solutions. Their relationships have been probed kinetically with proteases and with detergents. Some membrane enzymes are involved indirectly in recognition phenomena by modifying ligands that interact with cell surface receptors. The enzyme, therefore, has the capacity to terminate and possibly to modulate the signal for increased ionic conductance. The evidence for and against the presence of glycosyltransferases at the external surface of plasma membranes and their possible role in recognition is discussed in the chapter. Some of the enzymes are involved directly in transport across membranes and some prepare their substrates for transport by converting them to products, which can be handled by other transport systems. (Na+ + K+)M2+-ATPase is the best characterized true transporting enzyme. Of the brush border hydrolases, sucrase, maltase, and possibly lactase are directly involved in transport, and aminopeptidase may be capable of directly carrying out the transport of amino acids.
