An optimal ratio of the rates of modification of uridine and cytidine residues is achieved at pH 10.0 and at a NH2OH concentration not higher than 10 M. Hydroxylaminolysis of uridine units proceeds as follows: uridine unit → ribosylurea unit → ribosyloxime unit. Modification of cytidine residues under these conditions proceeds as a pseudo-single-step process. The rate constants of these reaction steps have been determined. This allowed the amounts of native and modified pyrimidine units in RNA after hydroxylaminolysis at pH 10.0 to be calculated and, moreover, the conditions (temperature and time of the reaction) to be selected for the preparation of polynucleotides with the preassigned ratio of modified units of defined structure." @default.
