- W1490270457 abstract "1101 Objectives Intracellular myocardial calcium relates glucose influx. To assess whether a calcium channel blocker such as verapamil could decrease 18F-FDG uptake by the myocardium in mice. Methods Experiments were conducted on 10 male C57BL/6J mice (group A, n = 5, 26.3-30.2 g ; group B, n = 5, 29.4-37.6 g). Mice were studied by 18F-FDG PET/CT under basal condition first and, on a subsequent day, after administration of verapamil. Verapamil, injected 1 hour prior to FDG administration (16.3 ± 1.4 MBq) at a dose of 1 mg/kg (group A) or 20 mg/kg (group B). Both verapamil and FDG were administered intraperitoneally in the conscious mice in non fasting condition. All studies were performed with a dedicated small animal PET/CT system (ARGUS-CT). For each mouse, post-verapamil PET images were co-registered with basal PET images. Volumetric region of interest (VOI) was drawn containing the whole left ventricle myocardium in the basal study. The same VOI was applied to the post-verapamil scan. The SUV was used to quantify the mean myocardium FDG uptake. To evaluate the effect of verapamil on the FDG uptake by myocardium, the relative variation coefficient (RV) was calculated as RV (%) = 100x(SUVpost-verapamil - SUVbasal)/SUVbasal. Results Data were reported as median (rank). Group A: the SUV mean for “basal scans” was 2.6 (1.6-4.1) and the SUV mean for the “post-verapamil scans” was 1.7 (1.1-2.9) (p=0.043). Group B: the SUV mean for “basal scans” was 1.6 (1.3-2.0) and the SUV mean for the “post-verapamil scans” was 1.0 (0.9-1.4) (p=0.043). The relative variation coefficient (RV) in group A was -31% (-5%, -50%) (p=0.043); RV in group B was -37% (-10%, -51%) (p=0.043). Conclusions In this animal model there was significant reduction of FDG uptake in myocardium following verapamil administration.
