The kinetics of RNA synthesis normally showed a rapid burst of synthesis followed by a slower linear rate of synthesis. A simple model is described to rationalize the data. Two generalizations are: (i) DNA's having the general composition d(pyrimidine)n·d(purine)n were transcribed asymmetrically; (ii) synthesis of RNA's which contain the codons UAG or UAA as a repeating unit stopped completely after about 15 minutes due to a non-dissociable ternary complex of DNA, RNA and RNA polymerase.
