- W1499022122 abstract "Insulin resistance is a major pathophysiologic abnormality in type 2 diabetes and is intricately involved in the development of not only hyperglycemia, but also dyslipidemia, hypertension, vasculopathy, and atherosclerosis. The thiazolidinediones are potent insulin sensitizers and through their PPARγ agonist effects, they not only improve insulin sensitivity and glycemic control, but also have potentially beneficial effects on lipid metabolism, vascular tone, and endothelial function. However, the widespread use of these agents has been limited by the development in some patients of undesirable side-effects including undue weight gain, bone loss, fluid retention, edema, precipitation of CHF, and a possible association with bladder cancer. Research is ongoing to make available tailored selective PPARγ agonists and dual PPARα/γ compounds, which through selective agonist effects have beneficial effects on glucose, lipid and vascular endothelial metabolism and through their partial agonist/selective antagonist actions, do not possess the unwanted side effects of the PPARγ agonists." @default.
