- W1500142243 abstract "Chromatography on methylated albumin columns of RNA's from Lentil roots, shows the presence of rapidly labelled RNA's tenaciously bound onto the column and which cannot be eluted by high NaCl molarities. Elution at 35°, with sodium dodecyl sulfate, gives two peaks (α and β). At 70°, a third elution peak (γ) is obtained. For short labelling periods, RNA's of the α peak are uridine rich (UMP/AMP approx. 1.2), heterodisperse and may have an important percentage of high-molecular-weight species. RNA's from the β peak are adenine rich (UMP/AMP approx. 0.4). Their modal sedimentation constant is about 12 S. The mean life of the uridine-rich RNA's of the α peak is lower than 1 h; that of the RNA's from the β peak is higher than 2 h. RNA's from α peak are essentially, or exclusively, of nuclear origin. On the other hand, polysomic messenger RNA's are present in the β peak. A close similitude exists between uridine-rich RNA's described in animal cells and the nuclear rapidly labelled RNA's of Lentil roots contained in the α peak. Short treatment (45 min) of Lentil roots with indoleacetic acid increases the amount of nuclear uridine-rich RNA's. A prolonged treatment (5 h) causes an increase in various rapidly labelled RNA's, probably polysomic messenger RNA's. Treatment of excised roots with indoleacetic acid leads, after a lag period of about 5 h, to the synthesis of two peroxydases associated with the ribosomes. The mean life of their messenger RNA's is about 2.5 h. La chromatographie sur colonnes de sérum-albumine méthylée, des RNAs extraits de racines de Lentille, permet de montrer la présence de RNAs à marquage rapide fortement fixés sur la colonne, et qui ne peuvent en être élués par des molarités élevées de NaCl. L'élution à 35° par le dodécyl-sulfate de sodium fournit deux pics (α et β). A 70°, on obtient un troisième pic (γ). Pour des temps courts de marquage, les RNAs du pic α sont riches en acide uridylique (UMP/AMP environ 1.2), hétérodisperses et peuvent posséder des constantes de sédimentation élevées. Les RNAs du pic β sont riches en acide adénylique (UMP/AMP environ 0.4). Leur constante de sédimentation modale est d'environ 12 S. La durée moyenne de vie des RNAs riches en acide uridylique du pic α est inférieure à une heure; celle des RNAs du pic β est supérieure à 2 h. Les RNAs du pic α sont essentiellement, ou exclusivement, d'origine nucléaire. Par contre, les RNAs messagers polysomiques sont contenus dans le pic β. Il existe une similitude étroite entre les RNAs riches en acide uridyliue décrits dans les cellules animales et les RNAs nucléaires à marquage rapide présents dans la racine de Lentille et contenus dans le pic α. Un traitement bref (45 min) des racines de Lentille par l'acide β-indolylacétique accroît la quantité de RNAs nucléaires riches en acide uridylique. Un traitement prolongé (5 h) entraîne un accroissement de la teneur des cellules en diverses catêgories de RNAs, probablement des RNAs messagers polysomiques. Le traitement, par l'acide β-indolylacétique, des racines excisées, entraîne la synthèse, après un temps de latence de 5 h, de deux peroxydases associées aux ribosomes. La durée moyenne de vie de leur RNA messager est d'environ 2.5 h." @default.
