- W1503397305 abstract "Eight different amino acid analogues, one with three isomers (monofluorophenylalanine) and one with two isomers (β-thienylalanine), have been given in the absence of the homologous amino acid to HeLa S-3 suspension cells in exponential phase of growth under strict cultural conditions to allow careful comparisons to be made of their effects on entry of cells into mitosis. The fluorophenylalanine and the β-thienylalanine analogues were capable of inhibiting progression of cells from G2 into mitosis. The degree of inhibition was dose dependent and was considerably greater at a culture temperature of 39.5 than at 37 °C. Some direct and some indirect evidence is presented with these and the other analogues to suggest that inhibition could be correlated with the amount of incorporation into cellular proteins. p-Fluorophenylalanine was the most inhibitory analogue. Canavanine and ethionine disturbed the G2 to M progression of HeLa cells, but their actions were distinctly different from those described above in that cytotoxicity was clearly apparent before cells stopped cycling. The role of Phe analogues is discussed in terms of the synthesis and utilisation of proteins in cells moving through G2 into mitosis." @default.
