- W1504936161 abstract "This chapter discusses the cloning cell cycle regulatory genes by transcomplementation in yeast. The identification and molecular cloning of cell cycle regulatory genes can be achieved through a variety of approaches. Cloning strategies based on sequence similarity to previously identified genes, antibody cross-reactivity, or in vitro assays of cell cycle functions combined with biochemical purification have all proved successful. In contrast, genetic approaches offer the possibility of gene isolation through direct selection of defined biological functions in vivo. Gene cloning by functional complementation, typically of temperature-sensitive cell cycle mutants has been achieved in a large number of studies of the budding yeast Saccharomyces cerevisiae and the fission yeast Schizosaccharomyces pombe. Transcomplementation between different yeast species can be achieved using genomic DNA libraries, but in most cases an appropriate cDNA library is required." @default.
