The purified enzyme had a high specific activity and was homogeneous as checked by ultracentrifugation and electrofocusing. It could be dissociated into subunits by cold-treatment or sodium dodecyl sulfate denaturation. The molecular weights of the two major and three minor subunits could be estimated by sodium dodecyl sulfate gel electrophoresis. The native enzyme had an isoelectric point of 5.2 while the cold-denatured enzyme showed three main bands focusing at pH 5.0, 5.2 and 5.4. Kinetic properties (Vm and Km (ATP)) have been compared for the soluble and membrane bound ATPase in presence of various anions. Inhibitory effects of Quercetin and other flavonoids have been tested in order to get an insight on the interaction between ATPase and its natural inhibitor. Une ATPase soluble (F1) a été purifiée à partir de mitochondries de cœur de porc. L'enzyme purifiée présente une forte activité spécifique et est homogène d'après son profil en ultracentrifugation et son comportement en focalisation isoélectrique. F1 se dissocie en sous-unités par traitement au froid ou par dénaturation en présence de dodécyl sulfate de sodium. Le poids moléculaire des deux sous unités principales et des trois sous unités mineures a été estimé par électrophorèse sur gel de polyacrylamide. L'enzyme native présente un point isoélectrique de 5,2 ; traitée par le froid, elle se divise en trois bandes principales ayant des points isoélectriques respectifs de 5,0, 5,2 et 5,4. Les propriétés cinétiques (Vm et Km (ATP) ont été comparées pour l'enzyme soluble et lié à la membrane en présence de différents anions. L'effet inhibiteur de la Quercétine et d'autres flavonoïdes a été testé dans le but d'obtenir une information sur la nature de l'interaction entre l'ATPase et son inhibiteur naturel." @default.
