- W150938526 abstract "Stage 17 chick embryo somites were cultured on nutrient agar according to Lash (4, 5) and on reconstituted collagen fibrils, and the effect of the substrate on chondrogenic differentiation was determined by following the onset of cartilage-specific type II collagen. It was found that exogenous collagen type I and type II collagen fibrils stimulate initially chondrogenic differentiation, whereas later in culture outgrowth of fibroblast like cells exceeded the development of cartilage. On agar cartilage formation started at 4 days in culture and surpassed fibrogenic and myogenic differentiation. In contrast to somite organ culture, chondrogenesis of dissociated limb bud mesodermal cells was mostly favoured in suspension culture on agar (17). In monolayer culture however collagen substrates stimulated type II collagen synthesis to levels of 40-50% of the total collagen synthesis compared to tissue culture plastic surfaces, which allowed only 10-20% of type II collagen synthesis. Fibrogenic differentiation, measured by type I collagen synthesis, was on collagen substrates impaired by comparison to plastic dishes, however favoured compared to cells which were kept in suspension on agar. The results suggest that type I and type II collagen stimulate chondrogenic differentiation in vitro. This may be an experimental model for the stimulation of somite chondrogenesis by the notochord. However in limb bud chondrogenesis cell interactions and matrix components other than collagen seem to be more important factors in chondrogenic differentiation." @default.
