- W1512682743 abstract "Rat growth plate cartilage is used as a test model for a cartilage fixation method that yields homogeneous preservation throughout tissue blocks and reproducible results at the light- and electron-microscopic levels. These results are achieved by the addition of ruthenium (III) hexammine trichloride (RHT), a highly purified cationic dye of low molecular weight and high positive charge density. In addition, reproducible results in cartilage fixation also depend on the exact control of certain chemical and physical parameters (osmolarity, temperature, pH, nature and concentration of buffer) in fixation media; their optimal ranges are presented. All chondrocytes throughout the various zones of the growth plate are preserved as fully expanded cells, i.e., their plasmalemmas contact the pericellular matrix without leaving an empty artificial lacunar space between. Cytoplasmic organelles are adequately preserved for cytomorphometric analyses in all cells throughout the different zones. The artifactual nature of intracellular vacuole formation in chondrocytes, which is a common problem of all available aldehydic fixation methods, is demonstrated." @default.
