- W15167899 abstract "Ethanol (E) inhibits contractions of the whole rat vas deferns (VD) induced by transmural nerve stimulation (TNS) and tolerance develops to this effect. In the rat VD, drug-induced responses are often different in the epididymal (EVD) and the prostatic (PVD) halves. This study investigated the effect of E and E tolerance on the response induced by TNS in EVD and PVD. The VD of control or ethanol tolerant rats were removed and bisected into EVD and PVD. The tissues were mounted between ring electrodes and equilibrated at 1 g tension for 2 hours. They were stimulated with supramaximal pulses at 0.1 Hz and 0.5 msec duration. After the response stabilized, E was added cumulatively. The IC50 for E was determine by linear regression analysis. E tolerance was induced by orally administering E twice daily for two days according to the method of Majchrowicz. In separate experiments prazosin and {alpha},{beta}methylene ATP (mATP) were utilized to define the neurotransmitters involved. E produced a dose-dependent inhibition of the TNS-induced contraction of PVD (IC50 = 264{plus minus}20 mM). In PVD from E treated rats the IC50 of E significantly increased to 395{plus minus}41 mM. In control EVD, E had a biphasic effect on TNS-inducedmore » contraction. Low doses potentiated the contractions while higher doses inhibited (IC50 = 666{plus minus}55 mM). In EVD from E treated rats the IC50 of E (581{plus minus}16 mM) was not significantly changed.
