- W1518476658 abstract "Summary The results obtained in our attempt to maintain and multiply the milk agent in vitro are strongly indicative that this has been accomplished. Gradual morphological changes were observed in organ cultures of embryonic mouse skin inoculated with R III milk. These changes were expressed by a hyperdevelopment of the epithelial structures, while connective and adipose tissues underwent degenerescence and necrosis. Extracts from these cultures induced characteristic mammary tumors in which electron microscopy could detect the typical particulate bodies that are usually associated with spontaneous neoplasms. The number of tumors induced in the agent-free C57BL strain of mice after four passages and 35 days in vitro cannot be satisfactorily explained by a transfer mechanism involving only a dilution of the primary inoculum, but by multiplication. The mammary tumor agent of the mouse was heat-labile; it was entirely neutralized when kept for 90 hours at the incubating temperature of 37° C., unless living tissues were present. The lag period between the inoculation and the appearance of the first tumor was not shortened by the transitory passage of the agent in vitro . Continuous roller-tube cultures of normal mammary epithelium from adult mice did not show any morphological changes following the introduction of the agent. Bio-assays revealed a maintenance of the factor without multiplication for at least 3 months, after which it was no longer detectable. A natural resistance of the adult tissues of the C57BL strain or fundamental modifications of their metabolism due to the artificial conditions of life in vitro could be the possible causes for this inhibition." @default.
