The molecules that are the subject of this chapter are known by several names: immunoadhesins, receptorbodies, and ligand traps being the most common. The chapter illustrates that the proteins are made by fusing, in frame, the cDNAs encoding the areas of interest of the receptor with one encoding the immunoglobulin Fc region and then transfecting that chimeric DNA into cells which make the fused protein. In addition to their solubility advantages, proteins modified and expressed in this manner have a number of other very useful features. The presence of an essentially normal immunoglobulin Fc domain conveys a number of useful properties. In addition, as the examples addressed in the chapter, immunoadhesins are versatile and powerful tools. Both their strengths and limitations arise from the same properties, which need to be kept in mind while considering their use. They are convenient, soluble forms of cell surface or secreted proteins. They contain an essentially normal Fc domain, which gives many of the same properties as antibodies. They can be “made to order” from any species, as long as one has the necessary facilities. In many ways, their properties complement those of traditional antibodies.
