- W152079671 abstract "An exciting approach in cancer molecular therapy is the ability to target the developing blood supply of the tumor. In contrast to several other molecular therapy approaches, antiangiogenic molecular therapy can readily access its target—the tumor vasculature. Numerous viral and nonviral vectors that target tumor angiogenesis have demonstrated antitumor efficacy in preclinical models in vivo. Molecular therapy strategies that inhibit proangiogenic growth factors such as vascular endothelial growth factor (VEGF) or its receptors include antisense methods, RNAzymes, DNAzymes, RNAi, or a dominant-negative protein encoded from a gene. These therapeutic molecules could be incorporated within mammalian expression vectors (plasmids or viruses) and/or delivered as small therapeutic polynucleotide molecules. Alternatively, tumor angiogenesis could be inhibited by in vivo expression of “endogenous” antiangiogenic proteins or peptides such as thrombospondin 1 or 2. In this chapter, we discuss the current status and strategies of antiangiogenic molecular therapy." @default.
