- W1530034981 abstract "Publisher Summary This chapter discusses various approaches that can be used to uncover the biochemical functions of type III effector proteins. The chapter also discusses their effects on eukaryotic signaling cascades. Before embarking on a method of analysis for identification of the effector's target, it is helpful to obtain some preliminary characteristics about the effector protein. First, the amino acid sequence of the effector should be studied. Second, in the case where the activity of an effector protein can be predicted by secondary structure analysis, mutants harboring mutations at critical residues can be generated. Third, the production of a recombinant form of the effector is required for many of the methods described and, therefore, knowing the expression profile (solubility, stability, yields) for a particular effector is essential information. Several different approaches can be used to introduce effector proteins into host cells to study their biological effects on host signaling cascades. The easiest and most biologically relevant way to accomplish this is by bacterial infection. However, for a number of reasons it is advisable to introduce effector proteins into host cells using one or more additional methods such as transfection or microinjection." @default.
