- W1534648506 abstract "Publisher Summary Radioligand-binding assays are a mainstay in the pharmacological analysis of receptor function. With these assays, the location, expression levels, and pharmacological specificity of a large number of G protein-coupled receptors (GPCRs) have been characterized, in many cases for the first time. The Sf9 expression system is most useful for binding studies where the receptor is needed to exist in an essentially purely uncoupled form in the absence of coexpressed G proteins or to couple to a single G protein without interference from others. Studies of agonist-induced signaling of GPCRs in Sf9 cells expressing the receptor alone indicate that these GPCRs can indeed interact functionally with endogenous insect G proteins. However, because activation assays require a lower ratio of G protein-to-receptor than the stabilization of high-affinity agonist binding in radioligand-binding studies, it is possible to see activation of G proteins under conditions where no high-affinity agonist binding is observed." @default.
