- W1544601788 abstract "Publisher Summary This chapter describes techniques that have been used to analyze the biogenesis of pili via the chaperone usher pathway and model systems to study host-pathogen interactions mediated by type 1 piliated E. coli. Among the earliest events in many bacterial infections are the molecular interactions that occur between the pathogen and host epithelial cells. These interactions are typically required for extracellular colonization or internalization to occur and involve a complex cascade of molecular crosstalk at the host-pathogen interface. The techniques described in this chapter give investigators insights into ways to characterize new pilus gene clusters and analyze the interactions of new adhesive organelles with host cells. A variety of biochemical, genetic, electron microscopic, and X-ray crystallographic techniques have been described to identify the function of each of the genes in a pilus gene cluster. Additionally methodologies are described to investigate adherence to the host and the consequences of this event using both in viva and in vitro model systems." @default.
