- W1546861808 abstract "This chapter examines the generation of DOTA-conjugated antibody fragments for radioimmunoimaging. The methods used for generation of radiolabeled monoclonal antibodies for use as imaging probes are presented. Antibody fragments may be preferable to intact antibodies as dynamic imaging probes due to their shorter biologic half-lives. As a result of their smaller size, they are cleared more quickly from nonantigen-expressing tissues and therefore provide greater tumor to normal tissue contrast at early time points. Conjugation of DOTA to intact antibodies or fragments involves first generation of the active ester of DOTA, which is then reacted with lysine residues on the protein. The conjugated antibody is then purified by ultrafiltration prior to radiolabeling. Using molecular imaging probes based upon conjugated antibody fragments, the expression of these targets can be noninvasively quantitated. This should allow for the real-time identification of patients, whose tumors express a particular molecular target, thus facilitating their inclusion in clinical trials of novel targeted agents." @default.
