It was found that spermine stimulated phenylalanine incorporation by a cell-free extract of Thermus thermophilus at the high, physiological temperature of this extreme thermophile. Without spermine the reaction stopped at an early stage, resulting in poor total incorporation. Addition of spermine increased the rate and the saturating level of phenylalanine incorporation at 50–80 °C. 2. Spermine and a native polyamine extracted from the thermophile were the most effective of the amines tested so far. Spermidine was less effective and other polyamines including putrescine had no effect on the incorporation. The optimum Mg2+ concentration was lowered to 6–10 mm by addition of 3 mm spermine. 3. Spermine was required in the formation of a ternary complex of 70S ribosomes, polyuridylic acid and phenylalanyl-tRNA and also for chain elongation in polyphenylalanine synthesis at high temperature. Putrescine inhibited the stimulating effect of spermine for the former step. Amino acyl-tRNA formation was not affected by the addition of polyamine. The action of the native polyamine extracted from the thermophile resembled that of spermine. 4. It was also found that preincubation in the cold enhanced polyuridylic acid-directed incorporation of phenylalanine by the thermophile extract without spermine at high temperature. The mechanism of enhancement by preincubation in the cold was compared with that by addition of spermine." @default.
