- W1550119510 abstract "The differentiation mechanisms that force thymocytes into the T-cell receptor (TCR)-αβ or TCR-γδ lineage are poorly understood, but rearrangement processes in the TCR-α/δ locus are likely to play an important role. It is assumed that the TCR-δ gene is deleted prior to Vα-Jα rearrangements by rearrangement of the so-called TCR-δ-deleting elements δRec and ψJα. However, the TCR-δ gene can also be deleted via Vδ-Jα rearrangements. We studied the different TCR-δ-deleting rearrangements of Vδ1, δRec, Vδ2 and Vδ3 to Jα gene segments in human thymocytes and peripheral blood using polymerase chain reaction analysis. The Vδ1 gene segment is the most upstream Vδ gene segment tested and appears to rearrange to almost all Jα gene segments. In contrast, the δRec and Vδ2 gene segments only rearrange to the 5′-located Jα gene segments, thereby preserving an extensive TCR-α combinatorial diversity, because most Jα gene segments are kept available for subsequent Vα-Jα rearrangements. Based on our combined data we hypothesize that the different Vδ gene segments and the δRec gene segment play different roles in T-cell development with regard to TCR-δ deletion." @default.
