- W1551130549 abstract "Cell transplantation is emerging as a novel strategy for myocardial regeneration, but it has been hampered by the lack of a source for human cardiomyocytes. Similarly, the lack of a human cardiomyocyte cell line has significantly limited a variety of experimental procedures. The recent advances in human embryonic stem cells (ES) research suggests a possible solution for this cell sourcing problem because these unique cells can be propagated in mass in vitro and coaxed to differentiate into the desired lineage. One of the most fascinating and important aspects of human ES cell lines is their ability to differentiate in vitro to advanced derivatives of all three germ layers. A prerequisite for this differentiation is the cultivation of the ES cells into three-dimensional cell aggregates. These aggregates, termed embryoid bodies (EBs) contain differentiating tissue from all three germ layers. Using the EB differentiation system cardiomyocytes can be reproducibly generated from the human ES cells. This chapter describes the methodologies associated with human ES cell cultivation and differentiation to cardiomyocytes, as well as the techniques used for studying the structural and functional properties of these unique cells." @default.
