- W1552336002 abstract "This chapter discusses procedures for using Ch as a mechanistic reporter, using photosensitization as a means of oxygen activation in two different test systems: isolated erythrocyte membranes and L1210 leukemia cells. Two techniques for analyzing 1O2-derived cholesterol hydroperoxides (ChOOHs) and distinguishing them from free radical-derived counterparts are described: (1) a relatively simple thin-layer chromatographic (TLC) method using colorimetric detection and (2) a state-of-the-art method involving high-performance liquid chromatography with mercury cathode electrochemical detection [HPLC-EC(Hg)]. By using these strategies, one can monitor the formation of 1O2, for example, in a biologically relevant milieu (membrane), thus avoiding the difficulties associated with external detection, e.g., by physical means. These approaches would be valuable for assessing reaction mechanisms for various oxidative agents of biomedical importance, including environmental phototoxins and the rapidly emerging family of phototherapeutic drugs.
