- W1554928752 abstract "Publisher Summary This chapter describes the procedures for preparation of basal and activated in vitro transcription systems for RNA polymerase II (Pol II)-dependent genes. The basal system also supports RNA polymerase I- and III-dependent transcription. Development of basal and activated plant in vitro transcription systems is critical for elucidating the biochemical mechanism of gene transcription and its regulation in plants. Tobacco nongreen cultured BY-2 cells are a good source for this purpose because this cell line has the fastest growth rate among the plant cell lines reported and the tobacco plant provides a powerful tool with which to analyze plant gene transcription in vivo by the well-established transgenic technique. As BY-2 cells do not differentiate in response to light or any other stimuli, they give a basal in vitro transcription system that lacks tissue-specific and light-responsive factors. By combination of the basal transcription system and active fractions from organs and tissues in which genes in question are specifically transcribed, cis-elements and trans-factors involved in their transcription are expected to be detected in vitro ." @default.
