- W1556111964 abstract "Functional analysis of mammalian striatin-family proteins gain in importance since they are thought to act as central components in calcium-dependent signal transduction pathways, which regulate cellular differentiation. In this process striatin proteins seem to provide a scaffold for the recruitment of large protein complexes, including amongst others kinases and phosphatases. One known interaction partner of mammalian striatin proteins is phocein/Mob3, a protein that seems to be involved in vesicular trafficking. The present work deals with the functional analysis of the fungal striatin and phocein/Mob3 homologs PRO11 and SmMOB3 from the filamentous ascomycete Sordaria macrospora. A yeast two-hybrid study revealed physical interaction of PRO11 and SmMOB3. Expression patterns of the corresponding genes pro11 and Smmob3 were analyzed using quantitative real-time PCR. An upregulation of both transcripts was observed during all, but especially increasing at late phase of sexua! l differentiation. Moreover, it was shown by deletion studies that the pro11 and Smmob3 genes are essential for fruiting-body development. Complementation assay identified the highly conserved 200 amino acid SmMOB3 Mob-domain as functional important protein structure. For PRO11 a functional correlation between specific protein domains and attained stages of fruiting-body development was shown. Furthermore, developmental defects of the deletion strains Δpro11 and ΔSmmob3 could not be complemented by crossing both strains with each other, indicating PRO11 and SmMOB3 to be present in a collective protein complex which mediates hyphal fusion. In a cross-species yeast two-hybrid screen of a Neurospora crassa cDNA library, NCU09375 was identified as an interaction partner of PRO11 and SmMOB3. The homolog gene Sm9375 was isolated from S. macrospora and functionally characterized. Sm9375 exhibits a putative N-terminal signaling sequence and a C-terminal ω-site which are both str! uctural characteristics of GPI-anchored proteins. In contrast ! to PRO11 and SmMOB3, Sm9375 is not essential for fruiting-body development but gene deletion resulted in a significant number reduction of produced perithecia. Moreover, additional deletion of Sm9375 in ΔSmmob3 partially complemented the ΔSmmob3 developmental defective phenotype, indicating a genetic interaction of the Sm9375 and Smmob3 genes." @default.
