Vibrational information is obtained that may be representative of one or more conformational states that are present at the same time. To obtain a Raman spectrum of a protein or nucleic acid, one places the sample at the focal point of a focused laser beam. Because a laser beam can be focused into a circle with a radius of approximately its wavelength, this allows for a spatial resolution of the order of a cubic micrometer. There are three types of Raman spectrometers in general use today for the characterization of protein or nucleic acid structure: (1) the classic laser Raman spectrometer, (2) the laser Raman microscope, and (3) the ultraviolet resonance Raman spectrometer. Classical Raman spectra of solutions of a protein or a nucleic acid are obtained from samples in a glass capillary mounted in a copper block with the temperature controlled by circulating water from a water bath." @default.
