- W1561691844 abstract "A new in vitro culture technique has been developed for the generation of cell-mediated immune responses of mouse spleen cells against cellular alloantigens. The system is modeled on a “one way” mixed lymphocyte reaction. Antigen recognition and subsequent cell proliferation are measured by 3H-thymidine uptake. The effector phase of the immune response is assayed by a highly sensitive 51Cr-release test, using tumor target cells, which share H-2-specificities with the immunizing cells. The culture system used is basically a modification of that described by Marbrook and is 30-fold superior to “classical” cultures in tubes. The modified system allows the culture of up to 60 × 106 immunocompetent mouse spleen cells for 6–7 days with a viability of 10–15%. This period of culture was required for the development of maximal cytotoxic activity. In vitro stimulated cells could lyse allogeneic target cells rapidly. At a 1001 ratio, 100% lysis always occurred within 200 min. A comparison of the activity of in vivo and in vitro stimulated cells showed an almost identical cytotoxicity, expressed both as rate of lysis and total lytic activity. The timing of the generation of cytotoxic activity was also very similar. Complement was not involved in the cell mediated lytic action as evidenced by the lack of effect of complement inhibitors. This in vitro culture system is highly reproducible (SD ± 5%) and the magnitude of the immune response is as great as that obtained with the most potent in vivo stimulation model known, namely the “one way” graft-versus-host-reaction." @default.
