- W1562704431 abstract "Abstract Tumor-associated fetal antigens (TAFA) are antigenic in syngeneic NZB rats and are immunogenic in 3-methylcholanthrene-induced (MCA) tumor-immune animals. Antibody-complement-dependent cytotoxicity tests demonstrated fetal specificities in serum from multiparous, MCA tumor-immune (but not MCA tumor bearer), and Moloney sarcoma virus Soehner-Dmochowski strain (osteosarcoma) tumor-bearing rats. Quantitative absorptions revealed antibody specificity for TAFA I (low-molecular-weight basic glycoprotein) in tumor-immune animals. Fluorescent antibody tests of sagittal sections of 13- or 15-day NZB fetuses were conducted to determine the normal tissue distribution of the purified TAFA. TAFA I is found only in areas of chondrogenic ossification. Since TAFA I is found in both fibrosarcomas and osteosarcomas, dedifferentiation to common bone and connective tissue stem cell equivalents is suggested. The cell distribution of fetal antigens in embryonic and transformed cells was determined, and membrane, cytoplasmic, and nuclear localities were found. Using syngeneic sera, it was found that the specificities for fetal antigens regarding cell location of fluorescent antibody activity is dependent on whether the animal was immune to tumors (fibrosarcomas) or tumor bearing. Although all sera tested gave membrane fluorescence, only MCA and Moloney sarcoma virus Soehner-Dmochowski strain tumor-bearing serum produced nuclear fluorescence. Additional cocapping experiments revealed that TAFA I is physically associated with NZB histocompatibility antigens. These experiments demonstrate that TAFA may be important in the development of tumor immunity, that these antigens are present in the embryonic homologous stem cell, and that certain fetal antigens are physically associated with rat histocompatibility antigens." @default.
