- W1565629412 abstract "Cytochemically selective detection of protein-bound sulphydryl groups has been achieved at the submicroscopic level with the monofunctional mercaptide-forming compounds, 1-(4-chloromercuriphenylazo)-naphthol-2 (“mercury orange”) and methyl mercury chloride. The use of p-chloromercuribenzoate as an alternate sulphydryl reagent is also explored and found to yield similar effects under certain conditions. These reagents were applied in numerous ways so as to determine, and ensure, favorable conditions of experimental procedure for submicroscopic cytological study. The reagents were used with due regard to securing optimal stoichiometric relations under non-oxidative conditions during reaction as well as during other stages of specimen preparation. The ultrastructural cytochemistry of sulphydryl groups is discussed in terms of protein denaturation, solubility, oxidation, enzyme digestion, and blocking reactions, including mercaptide blockage, which were employed to establish the specificity of localization. The morphological observations include the disclosure of an integral, -SH rich moiety of yeast ribosomes. These, and other observations are discussed in the light of independent data from the analytical chemistry and cytology of yeast." @default.
