They react with penicillin to form a covalent linkage, believed to be a thioester bond between a cysteine residue in the protein and the carbonyl of the β-lactam ring of the penicillin. The penicilloyl-enzyme bond can be cleaved by mild treatment with neutral hydroxylamine. Such treatment, moreover, restores enzymatic activity, at least in the case of the D-alanine carboxypeptidase of Bacillus subtilis. The above findings suggested the feasibility of covalent affinity chromatography as a means of purifying penicillin binding proteins. For preparation of the penicillin derivative, 10 ml of packed succinyldiaminodipropylamino-Sepharose is suspended in water to yield a final volume of 15 ml. Dimethylformamide (15 ml) is added, and the mixture is cooled to room temperature. The pH is adjusted to 4.8 with 1 N HC1, and 0.35 mmole of 6-aminopenicillanic acid (Sigma) is added. Next, 2.5 mmole of 1-ethyl-3(3-dimethylaminopropyl) carbodiimide dissolved in 1 ml of water is added dropwise over a 5-minute period. The pH is maintained at 4.8 for 1 hour by titration with 0.1 N HCl. The reaction mixture is gently stirred overnight at room temperature." @default.
